Laser ultrasonics measurements on Multicellular Tumor Spheroids

Résumé

Multicellular Tumor Spheroids (MCTS) are a powerful tool to investigate the mechanics and biology of tumors. MCTS are aggregates of cancer cells that mimic the 3D structure of avascularized tumors, with a structure that can be recapitulated as an elastic meshwork of actin bathed in biological liquids. This structure results in a complex mechanical behavior which is often described by analogy with the theory of poroelasticity. However, no strong proof of such a behavior has been observed in the past. Here we study the mechanical properties of spheroids in the MHz range with Laser Ultrasonics(LU). In the LU setup, we place a spherical MCTS of diameter 300 �m atop a 3mm aluminum plate. We place a semi-reflective glass blade on top of the MCTS is then compressed between the two plates. The whole setup is placed in a thermalized well filled with culture medium. We shine a pulsed laser on the bottom surface of the bottom plate, and the thermoelastic relaxation launches a broadband acoustic pulse that propagates through the plate, into the MCTS, and reaches the top reflective blade. The reflective blade is used as a mirror to detect the normal displacement with a stabilized Michelson interferometer. The blade is partially transparent to allow monitoring of the diameter of the MCTS during the experiment. Time-of-flight measurements allow accessing the sound velocity in a 1-100MHz frequency range. We engineered MCTS from two colorectal cancer cell lines. We measured their sound velocities while performing quasi-static compression. We observed different sound velocities for the two cell lines, paving the way for applications in tumor grading. In addition, comparison of the measured diameter with theory suggests expulsion of cellular fluids during the compression. This information, together with the sound velocity, brings new information on the poroelastic behavior of MCTS.
